甘肃河西地区西门塔尔杂交类群NGB基因第4外显子的遗传变异研究

Genetic Variation Study of NGB Gene Exon 4 of Simmental Cattle Hybrid Taxon in Gansu Hexi Regions

摘要: 为研究甘肃河西的临泽、甘州、凉州、金昌、高台5个地区西门塔尔杂交类群 NGB 基因第4外显子的遗传多态性及其变异特征,采用PCR-SSCP方法检测了283头西门塔尔杂交类群 NGB 基因第4外显子和部分内含子的多态性,并对群体内各等位基因进行了测序。结果显示:5个地区西门塔尔杂交类群共检测出5个等位基因(A、B、C、D、E),表现为6种基因型(AA、BB、AB、AC、AD、AE)。5个地区西门塔尔杂交类群均能检测到AA、BB、AB 3种基因型,且甘州、凉州、高台西门塔尔杂交类群还分别检测到AC、AD、AE 3种基因型。A等位基因频率在5个群体中最高,为优势等位基因。对不同SSCP带型的对应片段进行测序分析,共发现5个核苷酸突变位点(31 bp C→T、68 bp G→C和G→A、129 bp A→G、207 bp C→T),5处突变中仅第207 bp处单核苷酸突变落在外显子区域,其余均落在内含子区域,但均未导致氨基酸发生改变。χ 2检验结果显示,5个地区西门塔尔杂交类群在第207 bp处单核苷酸突变位点上仅凉州、金昌西门塔尔杂交类群处于Hardy-Weinberg平衡状态(P > 0.05)。群体遗传学分析结果表明,临泽、甘州、凉州、金昌、高台西门塔尔杂交类群的多态信息含量(PIC)分别为0.358 2,0.383 9,0.427 9,0.368 2,0.394 7,均属于中度多态(0.25

Abstract: The study was designed to analyze the genetic polymorphism and variability of NGB gene exon 4 of Simmental cattle hybrid taxon in Gansu Linze, Ganzhou, Liangzhou, Jinchang, and Gaotai five regions.DNA fragment containing the exon 4 and part of intron of NGB gene were amplified and genotyped using PCR single-strand conformational polymorphism (SSCP) method.Representative alleles were sequenced for verification of their variations in DNA sequences.The results displayed that there were 5 alleles (A, B, C, D and E) detected, combined six genotypes as AA, BB, AB, AC, AD and AE.Among them, AA, BB and AB genotypes were all detected in the five regions.AC, AD and AE genotypes was detected in Ganzhou, Liangzhou and Gaotai respectively.Allele A was the predominant genotype in five regions.Sequence of the SSCP showed 5 mutations in the DNA fragment (31 bp C→T, 68 bp G→C and G→A, 129 bp A→G, 207 bp C→T).In which, C→T at 207 bp was located in exon, while the other mutations were located in intron.But all had not leaded to amino acid changed.Chi-square testing indicated that in the 207 bp single nucleotide mutation sites, only Liangzhou, Jinchang Simmental cattle hybrid taxon among the five regions were in Hardy-Weinberg equilibrium (P>0.05).Population genetics analysis showed the polymorphism information content were 0.358 2, 0.383 9, 0.427 9, 0.368 2 and 0.394 7 in Linze, Ganzhou, Liangzhou, Jinchang, Gaotai, respectively, which were all at a moderate level of polymorphism (0.25
