PPARα基因对安卡×固始鸡资源群胴体品质的遗传效应分析

Genetic Effects of PPARtr Gene on Carcass Traits in Anak×Gushi Chicken Resource Population

摘要: 由793只安卡×固始鸡F2资源群为试验材料,采用PCR-SSCP技术和DNA测序的方法对PPARα基因的编码区进行SNP检测。结果发现,外显子4存在1个多态性位点,经限制性内切酶BspHⅠ酶切后显示出EE、EF、FF 3种基因型。利用SAS8.0软件分析不同基因型对胴体品质的影响,结果表明,PPARα基因NC006088:T32311C的多态性显著影响鸡的腹脂重和腹脂率(P<0.05),而对屠体重、皮下脂肪厚、肌间脂肪带宽、半净膛重和全净膛重等无显著影响(P>0.05)。EE型和EF型的腹脂重和腹脂率显著高于FF型(P<0.05),而EE型和EF型之间无显著差异(P>0.05)。PPARα基因影响脂肪代谢,可以作为分子标记用于鸡脂肪性状的分子标记辅助选择。

Abstract: A Anak×Gush resource population including 793 F2 chickens was founded by F2 design to analyze the PPARa gene polymorphism by the PCR-SSCP combined with sequencing techniques.The single nucleotide polymorphism(SNP)were found in exon 4 regions of the PPARa.Three kinds of genotypes detected in the resource population were defined as EE, EF and FF by digesting with BspH I restriction endonuclease.Effects of different geno.types on carcass traits were evaluated with SAS8.0 software.Results showed that the polymorphism of loci NC-006088:T32311C in PPARα gene was significantly related to the abdominal fat weight(AFW)and abdominal fat percentage(AFP)(P<0.05), and no significance with carcass weight, subcutaneous fat thickness, fat band width, semi—evisceration weight and evisceration weight(P>0.05).The chickens with the genotype of EE and EF had the higher AFW and AFP than those of FF(P<0.05).while there was no significant difference between the genotype of EE and EF(P>0.05).Result implied that PPARa gene affecting lipometabolism was a good marker for using in marker associated selection program.
