60Co γ射线辐照花生种子后代的SSR分析

Microsatellite Analysis on Mutants of Peanut Induced by 60Co γ

摘要: 以辐照处理花生品种鲁花11号M2为材料,通过农艺性状调查筛选变异材料。利用107对SSR引物对7个有代表性的变异材料进行PCR扩增,分析花生辐照诱变材料的SSR遗传多态性。同时将筛选到的多态性标记用于分析原品种和诱变后代材料间的遗传相似性。结果表明,诱变材料与原品种之间存在不同程度的多态性,且呈现多个位点突变;60Co辐照诱变可使花生的DNA分子多个区段内发生重复或缺失等结构性变异,并非单纯的点突变。诱变后代材料的成对相似系数变幅为0.457~1.000,平均值为0.862;诱变后代材料与其未辐射处理对照的遗传相似系数变幅0.692~1.000,平均值为0.873。由此可见辐射可创造丰富的遗传变异,拓宽原品种的遗传基础。

Abstract: Mutagenic progenies M2, selected from peanut variety Luhua11 irritated by γ ray of 60Co,had significated difference on leaf characters and plant types.The DNA polymophisms of seven mutants were analysed by using 107 pairs of peanut SSR primers.The polymorphism was various among mutant materials and origional variety Luhua11.Mutant materials had more than two different loci from origional variety.Polymorphic analysis of SSR marker indicated that mutation can be induced by γ ray of 60Co,mainly showing chromosome deficiency and chromosome duplication.The genetic similarity( GS) among 101 mutagenic progenies M2 were calculated based on the data from SSR markers.The value of GS ranged from 0.457 to 1.000,with an average of 0.862.The variance ranges of genetic similarity of mutagenic progeny M2 and their original variety Luhua11 were 0.692-1.000.The analysis of molecular markers indicated that abundant genetic variations can result and genetic bases in original varieties can be broadened from the γ-ray radiation.
