小麦黄花叶病毒河南驻马店分离物的鉴定与全序列分析

Detective and Complete Sequence Analysis of Wheat Yellow Mosaic Virus from Zhumadian in Henan Province

摘要: 采集河南省驻马店市13个不同田块小麦样品,利用RT-PCR、ELISA和Western Blotting检测技术进行检测,结果表明,该地有小麦黄花叶病毒的存在与分布。对检测为阳性的一个分离物全序列进行cDNA克隆和测序,与已报道WYMV河南潢川、江苏扬州、四川雅安和日本分离物的全序列进行比对,RNA1核苷酸一致性为97.1%~98.4%;RNA2为95.1%~98.4%;氨基酸一致性分别为94.8%~97.8%和94.2%~98.2%。利用软件MEGA4.1对上述五个RNA1和RNA2的全序列分别进行进化树分析,结果表明,驻马店分离物与潢川亲缘关系最近,而与四川雅安亲缘关系最远。

Abstract: A total of 13 wheat samples from different fields in Zhumadian of Henan Province were investigated,using RT-PCR,ELISA and Western Blotting,the results showed that WYMV existed there.Complete RNA genomic cDNA of one positive isolate(WYMV-ZMD)was cloned and sequenced,and was compared with those of isolates from Huangchuan,Yangzhou,Ya'an and Japan.Sequence comparisons showed that WYMV-ZMD shared 97.1%-98.4% nucleotide sequence identity for RNA1 with the reported isolates,and 94.2%-98.2% identity for RNA2.At the amino acid level,WYMV-ZMD had 94.8%-97.8% identity for RNA1,and 94.2%-98.2% identity for RNA2,respectively.Further phylogenetic analysis of five different isolates based on RNA1 and RNA2 by MEGA 4.1 indicated that WYMV-ZMD was more closely related to WYMV-HC,and had a more distant relationship with Ya'an isolate.
