钙与钙调素对番茄果实乙烯生物合成和信号转导基因表达的调控

Effect of Calcium and Calmodulin on the Expression of Genes in Ethylene Biosynthesis and Ethylene Signal Transduction

摘要: 为了研究CaM和Ca2+在调控乙烯生物合成和信号转导组分基因表达中的相互关系,利用272 mmol/L CaCl2及272 mmol/L CaCl2+100 µmol/L W5(或W7)处理绿熟期番茄果实圆片,然后采用无菌培养的方法在20℃的条件下进行培养。结果表明:CaCl2处理能够抑制番茄果实圆片的乙烯生成,抑制LeACO1,NR,LeERF2基因的表达, 而CaCl2+W5(或W7)能消除Ca2+对果实圆片乙烯生成LeACO1,NR,LeERF2基因的表达的抑制作用,但CaCl2和 CaCl2+W5(或W7)处理均对LeACS2基因表达影响不明显。以上研究结果表明外源钙处理对乙烯生物合成和信号 转导途径中相关基因表达的影响可能与CaM有关。

Abstract: In order to address the role that calmodulin plays in the regulation mechanism of calcium on the genes expression of ethylene biosynthesis and signal transduction pathway, the studies were carried out. Discs excised from tomato fruit(Lycopersicon esculentum cv. Lichun)at mature green stage were treated with 272 mmol/L CaCl2 or 272 mmol/L CaCl2+100 µmol/L WS/W7 solution,and then cultured under the aseptic conditions at 20℃.The results showed that both ethylene production and expression of LeAC01,NR, LeERF2 genes could be inhibited by CaC12 treatment,and CaCI2+WS/W7 treatment could prevent the inhibitory effect of CaCI2 treatment on ethylene production and expression of LeAC01, NR,LeERF2 genes. However, either CaC12 or CaC12+WS/W7 treatment has little effect on the expression of LeACS2 gene. These indicated that the inhibitory effect of CaC12 on expression of genes in ethylene biosynthesis and ethylene signal transduction pathway might be related to calmodulin.
