VB1和干燥处理对小麦幼胚愈伤组织培养的影响

Effects of VB1 and Desiccation on Callus Culture of Wheat Immature Embryos

摘要: 以1个小麦品种为试验材料,研究不同浓度维生素B1和分化前的干燥处理对小麦幼胚愈伤组织诱导、分化过程的影响.结果表明,小麦幼胚愈伤组织的分化率在不同基因型之间差异显著,其中最高的为郑新992(71.9%);在愈伤组织诱导、继代以及分化培养的MS培养基中添加维生素B1(盐酸硫胺素)10 mg/L有利于小麦幼胚胚性愈伤组织的发生,同时对促进小麦幼胚愈伤组织的分化也有显著影响;在转入分化培养基前对愈伤组织进行干燥处理(12 h)可有效提高其分化频率.

Abstract: The effects of VB_(1) and desiccation on callus induction and bud differentiation of immature embryo from 14 wheat genotypes were studied.Result indicated:there was a significant difference between the various genotypes in bud differentiation frequency,and ZhengXin 992 was the highest(71.9%).VB_(1)(10 mg/L) supplemented into the induction?subculture and differentiation media can improved the embryogenic callus development and increased the bud differentiation frequency.12 h desiccation before differentiation culture improved the bud differentiation frequency of calli.
