外源DNA导入小麦后代变异株系的酯酶同工酶分析

A Study on the Esterase Isoenzyme of the Variants Derived from Barley DNA Introduction into Common Wheat

摘要: 将抗白粉病的大麦DNA导入感病的小麦品种后,获得了抗白粉病变异后代,对其中抗性稳定的5个(D2代)株系子粒酯酶同工酶(EST)进行了分析。结果,在变异株系的EST酶谱中,检测到供体大麦所具有的酶带(即转基因酶带)和新的杂种酶带,从而证实了大麦基因(或DNA片段)导入到受体中,并被受体细胞基因组整合与表达。

Abstract: Five stable plant-lines from resistance variants in D2 generation derived from introducing DNA of two-rowed barley (H. vulgare ssp. distichon Hsu) with resistance to powdery mildew into susceptable common wheat `Hua 76'via the pollen-tube pathway were analysed with esterase isoenzyme (EST). The results of EST detection showed that the lines increased one band which was present in the donor but was absent in the receptor, named as transgene enzyme band. Thus it is confirmed that barley genes (or DNA segments) were integrated into or recombined with receptor's wheat genome and expressed. The effects of exogenous DNA introduced into recipient on variation of the character and change of the isoenzyme activity in the progeny were also discussed.
