摘要: 以玉米愈伤组织为受体,借助GUS基因的短暂表达,优化了PDS-1000/He型基因枪和自制基因枪的基因枪转化参数。PDS-1000/He型基因枪转化玉米愈伤组织的最佳轰击参数为1350psi压力,25 In·Hg真空度,6cm射击距离。自制基因枪转化玉米愈伤组织的最佳轰击参数为0.6 MPa,10cm射击距离。

Abstract: Using maize calli as recipient cell,transforming parameters of PDS 1000/He gene delivery system and self made gene gun have been studied through GUS transient expression.The most suitable delivery parameters for PDS 1000/He gene delivery system are 1350psi of pressure,25 In·Hg of vacuum degree and 6 cm shoot distance of microcarrier;and for self made gene gun are 0.6 MPa pressure,10 c shoot distance of microcarrier.
