摘要: 旨在探索小麦叶锈菌寄主体外萌发的适宜载体及温度,并为小麦叶锈菌的孢子萌发阶段RNA转录组分析提供优良的试验材料。采用高湿度黑暗培养法,以新鲜小麦叶锈菌致病类型FHJT的夏孢子为供试材料,测试不同载体对小麦叶锈菌夏孢子萌发特性的影响。在所测最适温度20 ℃下,以6种不同材料为培养载体,小麦叶锈菌夏孢子的芽管生长率和芽管生长长度均有显著差异,在无纺布上的夏孢子的芽管长度最长达到465.85 μm,微孔滤膜上的夏孢子芽管伸长速度最快,而在这6种材料上夏孢子12 h的萌发率基本相同。微孔滤膜更利于萌发孢子的收集,因此,微孔滤膜是最适宜的萌发载体。

Abstract: The objective is to obtain the optimum culture substrates and temperature for artificial germination of Puccinia triticina and afford good experimental materials for RNA-seq of P.triticina.We cultured the fresh FHJT urediniospores in dark and high humidity to perform the effects of six different culture substrates on germination of it.The result showed that there was significant difference in germination ratio and length of urediniospore germ tubes on the six culture substrates under optimum culture temperature 20 ℃.The longest tube that germinated on nonwovens was up to 465.85 μm.However,germ tubes on microfiltration membrane grew faster than that on others,while there was no significant difference in germination ratio on these six culture substrates after germination started 12 hours.Microfiltration is the optimum culture substrates for germination of urediniospores of P.triticina.
