四川白鹅,i>OAZ1和OAZ2基因组织差异表达的研究

Study on Differential Expression of OAZ1 and OAZ2 Gene in Various Tissues of the Sichuan White Goose

摘要: 为阐明四川白鹅不同组织中OAZ1OAZ2基因表达规律,应用实时荧光定量PCR技术检测四川白鹅13种组织中OAZ1OAZ2基因相对表达量,分析不同组织中OAZ1表达量与OAZ2表达量比值(OAZ1/OAZ2)的变化规律。结果表明,四川白鹅大脑和脾脏中OAZ1高度表达,分别是心脏的8.74,8.45倍(P <0.05);胸肌、腿肌和卵巢组织中表达量最低,仅为心脏的0.15,0.42,0.46倍。视网膜中OAZ2高度表达,是心脏的6.79倍(P <0.05);肝脏、胸肌和腿肌组织中表达量最低,分别是心脏的0.47,0.08,0.19倍(P <0.05)。四川白鹅胸肌和腿肌中OAZ2表达占优势,其余各组织中OAZ1表达占优势。卵巢组织中OAZ1/OAZ2的值最高,其次是小脑组织,分别为26.62和17.72;胸肌和腿肌中OAZ1/OAZ2最低,分别为0.33和0.75。四川白鹅OAZ1OAZ2基因表达具有组织特异性,不同组织中OAZ1/OAZ2值不同,提示不同组织中OAZ1OAZ2的调控功能可能存在差异。

Abstract: The objective of this work was to elucidate the expression profiling of OAZ1 and OAZ2 in various tissues of Sichuan white geese. The relative abundance of OAZ1 and OAZ2 mRNA in thirteen tissues of Sichuan white geese were determined by quantitative real-time PCR. The ratio between the amounts of OAZ1 and OAZ2 gene expression was assessed. OAZ1 mRNA was most highly expression in brain and spleen,8. 74-fold and 8. 45-fold,respectively,compared with that in heart(P <0. 05). The amount of OAZ1 gene expression in breast,thigh muscle and ovarian tissues was lower,0. 15-fold,0. 42-fold and 0. 46-fold,respectively,compared with that in heart. The level of OAZ2 mRNA expression in retina was higher 6. 79-fold compared with that in heart(P <0. 05). The relative abundance of OAZ2 in liver,breast and thigh muscle was lower,0. 47-fold,0. 08-fold and 0. 19-fold,respectively,compared with that in heart(P <0. 05). OAZ2 mRNA was more abundant than that of OAZ1 in breast and thigh muscle. OAZ1 mRNA was more abundant than that of OAZ2 in other determined tissues in the experiment. The highest OAZ1 / OAZ2 ratio was found in ovary and cerebellum(26. 62 and 17. 72,respectively),whereas the lowest one was in breast and thigh muscle(0. 33 and 0. 75,respectively). OAZ1 and OAZ2 mRNA are ubiquitously and tissue-specificity expressed. The OAZ1 / OAZ2 ratio is dependent on the different tissue of Sichuan white goose. OAZ1 and OAZ2 may exert differently regulatory function in various tissues.
