HIF-1α基因G901A多态性与高海拔低氧适应的相关性

Association between the G901A PolymoRphism of HIF-1α Gene and Adaptation to High-altitude Hypoxia

摘要: 为了研究低氧诱导因子-1α(HIF-1α)基因与高海拔低氧适应的相关性,使用高分辨率熔解曲线分析(HRM)小片段法分型技术对112头藏羊(海拔高于3 000 m)和117头低海拔地区羊(海拔低于500 m)HIF-1α基因的G901A位点进行基因分型和分析。结果发现:HIF-1α基因的G901A位点在2个群体中均处Hardy-Weinberg平衡,基因型频率在高海拔和低海拔群体间均具有统计学差异(P<0.01)。其中在该位点G/G、G/A、A/A这3种基因型的构成在藏羊上分别为41.07%,46.43%,12.5%,而低海拔群体为66.67%,27.35%,5.98%,藏羊等位基因G、A的分布频率为64.29%和35.71%,而低海拔群体为81.34%,19.66%,HIF-1α基因在G901A位点的基因型和等位基因频率在2组之间比较差异有统计学意义(P<0.01)。研究结果显示,HIF-1α基因的G901A位点多态性与高海拔低氧适应相关,AA基因型可能更有利于低氧环境。

Abstract: To investigate the association between the G 901 A polymorphism of HIF-1αgene and adaptation to high-altitude hypoxia, the G901 A of HIF-1αgene were genotyped using high resolution melting analysis in 112 un-related healthy Tibetan sheep ( Higher than 3 000 m altitude ) and 117 low altitude sheep ( Lower than 500 m alti-tude).Our results showed all the genotypes of the loci were significantly different between the two groups ( P <0.01).The frequencies of genotypes G/G,G/A and A/A at HIF-1α901 locus in Tibetan sheep were 41.07%, 46.43% and 12.5%,respectively in low altitude sheep, they were 66.67%,27.35%and 5.98%.Whilst in Tibet-an sheep allele frequencies of G and A were 64.29%and 35.71%,respectively but they were 81.34%and 19.66%respectively in the low altitude sheep (P<0.01).Our results indicate that the polymorphism of genotype in the G901 A of HIF-1αgene were associated with adaptation to high-altitude hypoxia.The sheep with AA genotype may be more conducive to hypoxia.
