牛Src基因三个内含子单核苷酸多态性

Single Nucleotide Polymorphisms(SNPs)within Three Introns of Src Gene in Bos taurus

摘要: 采用巢式PCR、DNA测序、创造酶切位点法PCR(Created restriction site-PCR,CRS-PCR)及PCR-RFLP(Poly-merase chain reaction-restriction fragment length polymorphism assay)方法,研究中国荷斯坦牛、鲁西黄牛及渤海黑牛的Src基因内含子6,8,9的单核苷酸多态性(SNPs),发现了14062(C/T)、17302(G/A)和18107(T/C)共3个SNPs,均为首次报道的位点。Src基因14062(C/T)、17302(G/A)和18107(T/C)3个位点在中国荷斯坦牛和鲁西黄牛两个群体中的优势等位基因相同,分别为C、G、C,其等位基因频率分别为0.671/0.647、0.583/0.640、0.558/0.577,而渤海黑牛在这三个位点的优势等位基因分别为T、G、C,其等位基因频率分别为0.525,0.913,0.763。经χ2适合性检验,中国荷斯坦牛、鲁西黄牛和渤海黑牛三个群体在14062(C/T)位点均达到Hardy-Weinberg平衡状态(P>0.05);在17302(G/A)位点则均未达到Hardy-Weinberg平衡状态(P<0.05);中国荷斯坦牛和鲁西黄牛在18107(T/C)位点均达到Hardy-Weinberg平衡状态(P>0.05),而渤海黑牛未达到Hardy-Weinberg平衡状态(P<0.05)。中国荷斯坦牛和鲁西黄牛在14062(C/T)和18107(T/C)位点均表现为中度多态(0.25

Abstract: The single nucleotide polymorphisms(SNPs)within the intron 6,8 and 9 of Src gene in Chinese Holstein cattle and Luxi Yellow cattle and Bohai Black cattle were investigated by nested-PCR,DNA sequencing technology,CRS-PCR(Created restriction site PCR)and PCR-RFLP(polymerase chain reaction-restriction fragment length polymorphism assay)method.Sequencing results showed that three SNPs were identified,which were 14062(C/T),17302(G/A)and 18107(T/C),respectively.In Chinese Holstein cattle and Luxi Yellow cattle,predominant allele were identical at 14062(C/T),17302(G/A)and 18107(T/C)site,which were C,G and C and the allelic frequencies were 0.671/0.647,0.583/0.640 and 0.558/0.577,respectively.However,the predominant allele of Bohai Black cattle at three SNPs site were T,G and C and the allelic frequencies were 0.525,0.913 and 0.763,respectively.Chi-square test indicated that the genotypic frequencies of 14062(C/T)site of Chinese Holstein cattle,Luxi Yellow cattle and Bohai Black cattle were in accordance with the Hardy-Weinberg equilibrium(P0.05),however,at 17302(G/A)site,the genotypic frequencies of all three population were out of the Hardy-Weinberg equilibrium(P0.05).The genotypic frequencies of 18107(T/C)locus in the Chinese Holsteins and Luxi Yellow cattle population are in Hardy-Weinberg equilibrium(P0.05),whereas being out of Hardy-Weinberg equilibrium for Bohai Black cattle(P0.05).The value of polymorphism information content indicated that three Chinese cattle breeds possessed intermediate genetic diversity for 14062(C/T),17302(G/A)and 18107(T/C)locus(0.25PIC0.5),except for Bohai Black cattle at locus 17302(G/A).
