固相萃取-反相高效液相色谱法测定大鼠血清中的甘氨胆酸

Determining the Glycocholeic Acid of Rats Blood Serum with SPE HPI, C

摘要: 通过建立血清中甘氨胆酸浓度的测定方法,旨在为观察甘氨胆酸在大鼠血清中浓度的变化规律做试验依据,为开发有价值的动物源性药物做前期探索。试验采用固相萃取-反相高效液相色谱法(SPE-HPLC),色谱柱为Waters Xterra RP18(150 mm×4.6 mm,5μm),流动相为乙腈-KH2PO4溶液,检测波长213 nm,柱温30℃,进样体积10μL。结果在0.156 25~10.0μg/mL范围内甘氨胆酸浓度与峰面积呈良好的线性关系,回归方程Y=39 579X+57 823,R2=0.998 4 以信噪比S/N=3为标准,最低检测浓度为0.02μg/mL,平均回收率98.49% 日内RSD 0.52%~2.00%,日间RSD 0.74%~4.98%。该方法具有简便、稳定、灵敏、准确、实用等特点,适用于血清中甘氨胆酸含量的测定分析。

Abstract: In this paper,determination method of concentrations of glycocholeic acid in rats blood serum is estab-lished by reversed phase high perfoimance liquid chmmatography(RP-HPLC),This study aims to understand the varia-dons of concentrations of glycocholeic acid in rats blood serum,provide a new method for Clinical Detection of gycochole-is acid,development useful drug from alnimal source. Uses the HPLC law,the chromatographic column is Waters XterraRP18 (150 mm X4. 6 mm,5μm),flowing for second grade nitrile- KH2PO4solution,examination wave length 213 nm,col-umn warm 30 ℃,specimen handling plume l0 L. The result the glycocholeic acid density and the peak area assumes thegood linear relations in 0. 156 25-10. 0 μg/ mL scope,regression equation Y=39 579 X+57 823,R2=0. 998 4; Takesignal-to-noise ratio S/ N=3 as the standard,the lowest examination density is 0. 02μg/mL, The average recovery was98. 49%;the RSD of innerday was 0. 52%-2. 00%,the RSD of infra day was 0. 74%-4. 98%.The method was sinrple,accurate,sensitive,feasible and suitable for analyzing glycocholeic acid concentration in blood serum.
