牛早期胚胎性别鉴定快速PCR方法的研究

Sex Identification of Bovine Embryo with PCR Methods

摘要: 根据公牛Y-染色体特异重复序列和公母牛共有的DNA序列,设计并合成PCR引物用于牛或牛胚胎性别鉴定,通过PCR反应对公母牛静脉血及牛胚胎进行性别鉴定,结果表明,母牛只能扩增出250 bp的内标基因片段,而公牛能同时扩增出131 bp的Y-染色体特异性基因片段和250 bp的内标基因片段。静脉血样与实际性别的符合率为100% 4~10个胚胎细胞的扩增也出现和静脉血相同的特异条带。这表明该检测方法准确、简便,具有较高的灵敏性和稳定性。

Abstract: Two pairs of specific primes were designed on the special sequences of Y chromosome and sharing DNAsequence of bull and a set of polymerize china reaction were used for identify the bull and cow blood samples andembryo. Results showed,only demonstrated a band of 250 by and bull demonstrated two band of 131 by by and 250bp, result`s of blood identification is completely in accordance with the actual sex type, 4 to 10 embryo cell also demon-strated same band model of blood sample, which shaved that this method of sex identification were effective andsensrtrve.
