摘要: BMP15基因主要在哺乳动物卵巢中表达,对卵泡的发育和分化起重要作用。根据其他物种BMP15基因的保守序列设计特异性引物,扩增蒙古羊的全长DNA序列。从蒙古羊的血液中提取总DNA,采用PCR技术扩增出蒙古羊BMP15DNA序列。将此片段克隆到PMD18-T载体中,经菌落PCR鉴定和DNA序列测定分析验证,证实所克隆序列BMP15为骨形态发生蛋白,符合骨形态发生蛋白基因的特征。序列分析表明,该DNA包含两个外显子序列和一个内含子序列以及两端的非翻译序列,全长6 642 bp,编码393个氨基酸,蒙古羊与牛的同源性最高(98.6%),与猪的同源性为90.5%。

Abstract: BMP15 has a important role to the development and differentiation of follicular and is specialized erpressed in mammalian ovary. Specific primers was designed based on conserver sequence of the other animals,firstly,totalDNA was extracted from blood of Mongolian sheep and the BMPIS DNA was amplified by PCR,The purified PCR.prod-uct was cloned into PMD18-T vector. The results demonstrated that the Mongolian sheep BMP15 is belong to the family ofbone moiphogenetic protein. On the basis of comparing the sequence to cow and pig,the homologes of nucleotide were98. 6%,90. 5%respectively.
