西葫芦未受精胚珠离体诱导植株再生的关键因素

Critical Factors Influence in vitro Plantlet Production of Unfertilized Ovule Culture in Summer Squash(Cucurbita pepo L.)

摘要: 开花当天的西葫芦未受精胚珠在含5%蔗糖和0.8%琼脂的N6基本培养基中附加0.5 mg/L NAA和1.0mg/L 2,4-D的诱导培养基上培养最有利于诱导植株再生;所得再生植株经气孔保卫细胞叶绿体计数法鉴定,60%为二倍体,其自交后代株系内高度整齐一致,证明所得二倍体再生植株是纯合的双单倍体,来源于单倍的胚囊细胞。

Abstract: The results showed that N6 basal medium supplemented with 5% sucrose,0.8% agar,0.5 mg/L NAA and 1.0 mg/L 2,4-D facilitate plantlet induction of unfertilized ovule culture in summer squash(Cucurbita pepo L.).Diploid plantlets,which account for 60% of the total plantlets by taking account of chloroplast number in guard cell were confirmed of pure lines through investigation of agronomy traits of selfing offsprings.That means the diploids were pure line and they originated from gynogenesis of haploid megaspore cells.
