云南松毛虫肠道产纤维素酶菌株的筛选鉴定及酶学性质

Screening and Identification of Cellulase-producing Strains from Intestinal Canal of Dendrolimu houi and Research on Cellulase Characterization

摘要: 从云南松毛虫肠道中分离筛选得到28株产纤维素酶的菌株。经生理生化试验观察和16S rRNA序列比对分析,初步鉴定这些产酶菌株属于肠杆菌属、芽孢杆菌属、克雷伯氏杆菌属和约克氏菌属。同时筛选出一株产酶能力较高的菌株DK4,鉴定为蜡状芽孢杆菌,并构建系统发育树。对该菌株产酶特性进行了初步研究,其酶反应的最适温度是35℃,最适pH值7.0,连续发酵70 h左右时纤维素酶活达到最高值19.24 U/mL。

Abstract: Twenty-eight Cellulase-producing strains were isolated from intestinal canal of Dendrolimu houi. Basing on biochemical, physiological characterization and 16S rRNA sequences blasting, these strains were classifiedinto Enterobacter sp. , Bacillus sp. , Klebsiella sp. and Yokenella sp. . Strain DK4 was found as a higher-yield strainfor cellulase in these strains. A phylogenetic tree was constructed with the published 16S rRNA sequences of relativebacteria species. In the phylogenetic tree, the strain DK4 was in the branch with the Bacillus cereus strain NAV-10( GU056811 . 1 ) . The highest cellulase activity of DK4 strain was 19. 24 U /mL when grown at 35℃ , pH 7. 0 for 70 h.
