摘要: 利用分子克隆技术获得了牦牛VEGF-A基因编码区序列,并采用生物信息学方法对该基因及其编码蛋白的基本理化性质、疏水性、信号肽、二级结构等方面进行了预测和分析。结果表明,牦牛VEGF—A基因包含一个573bp的开放阅读框,编码190个氨基酸;其编码蛋白属于亲水性蛋白,具有明显的信号肽。二级结构主要以无规则卷曲和α螺旋为主。VEGF—A基因编码产物氨基酸邻接系统树表明,牦牛VEGF—A与黄牛、绵羊和成都麻羊等物种的VEGF—A氨基酸遗传距离较近,具有高度同源性。

Abstract: A coding region sequence of yak VEGF-A was cloned by molecular cloning technique in this study. Some characters of the VEGF-A gene and encoded protein sequences were predicted and analyzed by the methods of bioinformatics in the following aspects as the general physical and chemical properties, hydrophobicity, signal pep- tide and secondary structure. Results showed that the full-length of yak VEGF-A contains a complete ORF(573 bp) encoding 190 amino acid. The protein encoded by yak VEGF-A has obvious signal peptide,it is a hydrophilic pro- tein. The secondary structures are mainly composed of Helix and Coli. The VEGF-A and related proteins of Bos grunnien,Bos taurus,Ovis aries,Capra hircus are close in the phylogenetic tree,which showed highly homology.
