农杆菌介导棉花基因转化的研究

Study on the Transformation of Cotton Mediated with Agrobacterium tumefacines

摘要: 以棉花无菌苗下胚轴为外植体,通过愈伤组织培养,使供试的20个棉花新品种(系)中19个培养出体细胞胚或再生植株。在此基础上,选用易获得再生植株的邯93-2、邯无2031、衡无89-30、C201、C312,利用农杆菌介导转化Bt基因,抗卡那霉素愈伤组织诱导率约为50%,其中约40%表现GUS阳性。从邯93-2GUS阳性愈伤中获得再生植株,其50%呈GUS阳性,且Bt基因的导入经Southern杂交验证。

Abstract: Hypocotyl used as explants,twenty varieties or lines were studied for plant regeneration or somatic embryoid.Nineteen of them were successful.According to the result,the varieties easy in plant regeneration Han 93-2,Hanwu 2031,Hengwu 89-30,Coker 201,Coker 312 were chosen for the transformation.Inoculating rate of Kanamycine resistant callus was about 50%,among them 40% was GUS positive.50% of plants regenerated from the GUS positive callus were GUS positive,and proved by Southern hybridization analysis.
