大豆中3个Dof转录因子的生物信息学及表达分析

Bioinformatics and Expression Analysis of Three Dof Transcription Factors in Soybean

摘要: 植物Dof转录因子在植物应对非生物胁迫过程中发挥重要作用。为探明Dof转录因子参与大豆对非生物胁迫的响应,对大豆中3个Dof转录因子(GmDof2.1GmDof3.1GmDof4.6)的基因序列进行生物信息学分析。3个Dof基因分别位于不同染色体上,它们所编码的蛋白序列长度为212~305个氨基酸残基,均具有1个保守的Dof结构域。3个Dof蛋白主要定位于细胞核中且含有不同数量的磷酸化位点。启动子序列分析表明,GmDof2.1启动子序列中含有3种与逆境和激素响应相关的元件(ARE、DRE1和MBS),GmDof3.1启动子序列中含有6种与逆境和激素响应相关的元件(ABRE、ARE、CGTCA-motif、TGACG-motif、W-box和WUN-motif),GmDof4.6启动子序列中含有7种与逆境和激素响应相关的元件(ABRE、ARE、CGTCA-motif、GARE-motif、MBS、TGACG-motif和WUN-motif)。实时荧光定量PCR结果显示,3个Dof基因均可不同程度的响应高盐、干旱、低温和高温胁迫。GmDof2.1GmDof3.1在大豆根中的表达量最高,GmDof4.6在大豆茎中的表达量最高。由此推测3个Dof转录因子可能在大豆应对非生物胁迫过程中发挥转录调控作用。

Abstract: Dof transcription factors play an important role in plant response to abiotic stress. In order to investigate the Dof transcription factors of soybean involved in the response to abiotic stress, the bioinformatics analysis were performed on three Dof genes(GmDof2.1,GmDof3.1 and GmDof4.6) from soybean, which were located on different chromosomes. The length of the three Dof proteins ranged from 212 to 305 amino acid residues, all of which had a conserved Dof domain. They were mainly located in the nucleus and contained different amounts of phosphorylation sites. Promoter sequence analysis showed that GmDof2.1 promoter contained three cis-acting elements related to stress and hormone(ARE, DRE1 and MBS), GmDof3.1 promoter contained six cis-acting elements related to stress and hormone(ABRE, ARE, CGTCA-motif, TGACA-motif, W-box and WUN-motif), and GmDof4.6 promoter contained seven cis-acting elements related to stress and hormone(ABRE, ARE, CGTCA-motif, GARE-motif, MBS, TGACG-motif and WUN-motif). The results of Real-time fluorescence quantitative PCR showed that all three Dof genes could respond to the stresses of high salt, drought, low temperature and high temperature in varying degrees. GmDof2.1 and GmDof3.1 had the highest expression in roots, and GmDof4.6 had the highest expression in stems. It was inferred that the three Dof transcription factors might play a transcriptional regulatory role in soybean response to abiotic stresses.
