不同菌根真菌对铁皮石斛穴盘苗生长及生根的影响

摘要:

为了探讨菌根真菌与铁皮石斛穴盘苗之间的共生关系,旨在筛选出影响穴盘苗生长的最佳优良共生真菌菌株。以几种不同的兰科菌根真菌和铁皮石斛穴盘苗为试验材料,进行接菌共生培养研究,比较不同菌株对接种共生穴盘苗的影响,结果表明:穴盘苗PS1、PS2、PS3处理的平均鲜质量增长率分别比对照高15.7,7.4,19.7百分点,与对照相比差异均达极显著水平(P<0.01);穴盘苗PS1、PS2、PS3 、PS5处理的根增长率分别比对照高44.4,43.6,30.2,33.4百分点,与对照相比较差异达极显著水平(P<0.01)。同属于瘤菌根菌属的D4菌株在整个培养过程中一直表现出抑制作用,D5菌株培养后期的促进作用比前期更明显。从叶绿素含量变化情况也可看出,除了D4以外其他菌株可明显增加叶绿素的形成,促进穴盘苗生长。筛选出的最佳优良菌株为D1菌株、D2菌株(未知)和D3菌株,它们对铁皮石斛穴盘苗生长有明显促进作用,即将应用于后续铁皮石斛组培苗的移栽驯化。

Abstract:

In order to explore the symbiotic relationship between mycorrhizal fungi and Dendrobium officinale plug seedlings, the aim was to screen out the best and excellent symbiotic fungal strains that affect the growth of the plug seedlings.Different Orchidaceae mycorrhizal fungi and plug seedlings of Dendrobium officinale were used as experimental materials to study the symbiotic relationship between mycorrhizal fungi and plug seedlings, so as to screen out the best symbiotic fungi strains that affect the growth of plug seedlings of Dendrobium officinale.The results showed that:The average fresh weight growth rates of PS1, PS2 and PS3 were 15.7, 7.4 and 19.7 percentage higher than those of the control group, and the differences were extremely significant (P<0.01);The root growth rate of the plug seedlings PS1, PS2, PS3 and PS5 was 44.4, 43.6, 30.2 and 33.4 percentage higher than that of the control group, the difference reached a very significant level (P<0.01).The D4 strain which belonged to the same genus of Epulorhiza sp., showed inhibitory effect during the entire culture process, and the promotion effect of the D5 strain was more obvious in the later stage than in the early stage.It can also be seen from the change of chlorophyll content that other strains except D4 could significantly increase the formation of chlorophyll and promote the growth of plug seedlings.The best excellent strain selected in this research experiment was D1 strain (Epulorhiza sp.), D2 strain (unknown) and D3 strain (Rhizoctonia sp.) significantly promoted the growth of Dendrobium officinale plug seedlings. This would soon be applied to the subsequent transplantation and domestication of tissue culture seedlings of Dendrobium officinale, and would also provide theoretical and practical reference for the efficient in vitro culture and propagation of embryos of other orchidaceous medicinal plants.
