响应面法优化甜瓜枯萎病拮抗菌F1发酵条件

Optimization of Fermentation Conditions of Antagonistic Bacteria F1 Against Muskmelon Fusarium Wilt by Response Surface Methodology

摘要: 为了找出甜瓜枯萎病拮抗菌F1 的最佳发酵条件,应用Plackett-Burman设计试验确定无机盐含量、温度、豆饼粉含量是影响F1菌株有效活菌数的主要影响因子.经最陡爬坡实验、响应面优化,建立数学模型且通过模型得到最优发酵条件,当接菌量为4%、初始pH值为7、摇床转速为200 r/min、玉米粉含量为30 g/L、豆饼粉含量为17.92 g/L、MgSO4 0.604 g/L、NaH2PO4 1.51 g/L、CaCl2·2H2O 0.302 g/L、K2HPO4 1.51 g/L、培养温度为30.0 ℃时,有效活菌数为1.507×109 cfu/mL.验证试验显示真实值为预测值的101.0%,说明该模型可用于指导F1在实际生产中发酵条件的优化.

Abstract: In order to find out the optimization of fermentation conditions of antagonistic bacteria F1 against muskmelon Fusarium wilt.Plackett-Burman design was employed to investigate the main factors affecting colony-forming units of strain F1 and confirmed that temperature,content of inorganic salt and bean powder were the main factors.After steepest ascent experiment,response surface analysis and mathematic modeling we obtained the optimum conditions of fermentation as following:4% of inoculation;initial pH value 7;200 r/min of rotating speed;the content of corn powder,bean powder,MgSO4,NaH2PO4,CaCl2 2H2O and K2HPO4 was 30,17.92,0.604,1.51,0.302,1.51 g/L respectively;30.0 ℃ of cultivation temperature.So the colony-forming units of strain F1 increased nearly 117 times to 1.507×109 cfu/mL and the experimental values was 101.0% times to the predict values.These results revealed that the predicted model was reliable and available for the optimization of fermentation conditions of F1 during production.
