灰斑古毒蛾核型多角病毒EcoR I-O 片段的克隆与生物信息学分析

Analysis of EcoR I-O Fragment from Orgyia ericae Nucleopolyhedrovirus

摘要: 分析了灰斑古毒蛾核型多角体病毒(Orgyia ericae nucleopolyhedrovirus,OrerNPV)EcoR I-O 片段分子生物学特征,为研究egt基因分子特性和杆状病毒遗传改良提供科学依据.克隆和分析了OrerNPV EcoR I-O 片段所包含的2个基因,并应用生物软件对OrerNPV蜕皮甾体尿苷二磷酸葡糖转移酶(egt)基因及其启动子进行了比对分析.结果表明OrerNPV EcoR I-O片段包含egt基因和Ol-129同源基因,在该区域中编码egt基因的开放阅读框(ORF)由1 539个核苷酸组成,可以编码512个氨基酸,预计蛋白质分子质量为57.9 kDa.OrerNPV和NPVs之间的同源性为4%~89%,与白斑天幕毛虫核型多角体病毒(Orgyia leucostigma NPV,OlNPV)egt基因同源性最高,达89%,与棉褐带卷叶蛾核型多角体病毒(Adoxophyes orana NPV,AoNPV)同源性最低为4%;而OrerNPV和 GVs 之间的同源性为 44%~55%,小于与NPVs之间的同源性;与昆虫UGT间同源性最低,与家蚕UGT间同源性为41%.EcoR I-O片段+M13端含有编码OlNPV ORF129(Ol-129)Ol-129同源基因ORF,由597 bp个核苷酸组成,在其起始密码子ATG上游-4,-24 nt处各有1个TATA box,其推测的氨基酸序列与Ol-129同源性最高达81%,与LdNPV ORF127同源性最低,为40%.明确了OrerNPV EcoR I-O 片段的序列特征,以及OrerNPV egt 基因特性.

Abstract: It is not specified of the molecular characterization on Orgyia ericae Nucleopolyhedrovirus(OrerNPV).Here,the cloned OrerNPV EcoR I-O fragment was analyzed,which the egt gene and homologues of Ol-129 gene were found in this fragment.The OrerNPV egt open reading frame was 1 539 nucleotides long that coded a peptide of 512 amino acids with a predicted molecular weight of 57.9 kDa.The peptide of OrerNPV egt shared a highest similarity of 89% to Orgyia leucostigma nucleopolyhedrovirus;while it shared the lowest similarity with Adoxophyes orana nucleopolyhedrovirus 46%.The homologues of Ol-129 gene amino acid residues in the M13+ terminal of OrerNPV had 89%-40% identities with homologous genes from other 10 NPVs.The result showed that characteristic of OrerNPV EcoR I-O fragment and OrerNPV egt gene.
