摘要: 利用随机扩增多态性DNA技术(RAPD)对加拿大披碱草和老芒麦及其杂种F1, F2做了比较分析。旨在用分子标记方法探索亲本加拿大披碱草与老芒麦及其远缘杂种后代的遗传关系, 为后期育种提供理论依据。结果表明: 两亲本加拿大披碱草和老芒麦之间的亲缘关系较近, 杂种F1主要表现为互补双亲的RAPD扩增带, 同时还丢失了双亲部分的RAPD扩增带, 杂种F1的多态性条带有偏向母本遗传的倾向; F1是真正的杂种; F2的多态性条带表明: F2与父母本的遗传距离相差不大, 略偏向母本。亲本与杂种F1, F2的RAPD扩增带表型均具有一定的差异, RAPD遗传标记可用于杂种鉴定和目标性状植株的检测。

Abstract: Elymus canadensis, E.sibiricen and their Hybrid F1, F2 were analyzed by using random amplified poly-morphic DNA(RAPD)markers. The results showed that the two parents Elymus canadensis and E. sibiricus had relatively closer relationships. The RAPD bands of the hybrid F1 were mainly mutual supplement RAPD bands of the parents; at the same time, some RAPD bands of the parents were lost, the RAPD types of hybrid F1 showed a tendency toward the maternal type. The RAPD bands of F2 showed that the geneticrelationships between F2 and their parents were almost the same, but it also showed a tendency toward the maternal type a bit. There were differences about RAPD types between parents and their hybrids; RAPD technique cenhe used to identify hybrid and to detect the plantswith object traits.
