摘要: 分离筛选具有高效解磷解钾效果的菌株并进行鉴定及培养条件优化。采用鉴别培养基进行菌株分离筛选,通过生理生化鉴定及16S rRNA基因序列分析进行菌种鉴定,并对其发酵培养基和培养条件进行正交试验确定最佳培养条件。筛选出高效解磷解钾菌株NX-11。测序结果表明,NX-11与Bacillus subtilis subsp.Subtilis亲缘关系接近。确定其最佳发酵培养基配方为:淀粉5.0%,黄豆饼粉1.5%,MgSO40.07%,NaCl 0.05%|最佳培养条件为:种龄20 h、装量30mL/250mL三角瓶、接种量4%、pH值8.0培养温度37℃、摇床转速200 r/min。

Abstract: Bacteria having the efficient activity on solubilizing phosphate and potassium have been isolated for its utilization as a biological fertilizer. The most efficient strain named NX-11 was chosen for further study. NX-11 was identified as Bacillus subtilis after a series of physiological and biochemical experiments,morphological observa-tion and 16S rRNAgene sequence analysis. The solubilization ability of NX-11 was tested in the condition of pure cul-ture. The results showed that the optimal medial components were composed of 5.0% starch,1.5% soybean cake powder,0.07%mgSO4,0.05% NaCl. The optimal culture conditions were at temperature 37℃ with initial pH 8. 0 and incubation time 20 h,inoculum's volume 40mL/L,medium volume 30mL in 250mL flask and 200 r/min.
