荠菜多聚半乳糖醛酸酶基因CbPG的分子克隆、序列分析和功能预测

摘要: 根据白菜花粉特异的多聚半乳糖醛酸酶(PG)基因BcMF2(EU181170)的全长序列设计特异引物,通过PCR扩增的方法从荠菜中克隆了一个新的BcMF2的同源基因CbPG。该基因与BcMF2在核酸水平上的相似性高达99%。CbPG基因DNA全长为1 563 bp,由4个外显子和3个内含子构成,外显子总长为1 263 bp,编码420个氨基酸。序列比对结果表明,该基因具有所有PG基因特有的4个保守结构域,并且与花粉中特异表达的PG基因聚为一类,表明该基因可能是与花粉发育相关的PG基因,在花粉萌发和花粉管伸长中起作用。

Abstract: In the paper, CbPG gene, homologous with the BcMF2 gene ( EUI81170 ) encoding pollen-specific polygalacturoase (PG) of Chinese cabbage,was cloned from Capsella bursapastoris ( L. ) Medic. by PCR amplication technique. The CbPG gene showed 99% overall DNA similarity with that of BcMF2. The gene consists of 1 563 bp encoding a protein of 420 amino acids, interrupted by three introns. Sequence alignment revealed that four domains which are highly conserved in the all plant and fungal PGs is present in CbPG. Phylogenetic analysis showed that CbPG fails into the category of clade-C,which includes PGs related to pollen development. These results test that the CbPG gene might also acts as pollen-specific polygalacturonase.
