摘要: 对影响黄瓜枯萎病菌AFLP技术体系的多种因素作了探讨,得到了1种适合于黄瓜枯萎病菌AFLP分析的优化体系。结果表明,用于酶切的基因组DNA以100 ng(25μL体系)为宜,预扩增产物的稀释倍数以20倍为宜。采用优化好的体系,以21个不同地区的黄瓜枯萎病菌菌株为试材,引物组合E15/M65在21个菌株中共扩增出稳定清晰的带纹452条,各菌株扩增带分布较均匀,扩增带最少的为13条,最多的32条。

Abstract: In this paper, some factors affecting the AFLP researching system of Fusarium oxysporium were studied and an advanced technical system was established. With the optimized system, the AFLP fingerprinting of 21 Fusarium oxysporium strains collected from different regions amplified by primer combination E15/M65 have been obtained, which had 452 clear bands.
