黄瓜AFLP反应体系的建立

Construction of AFLP Analysis System in Cucumber

摘要: 以24个不同类型的黄瓜高代自交系为试材,通过对扩增长度多态性(AFLP)反应体系中的几个关键参数进行优化,建立了适合于黄瓜的AFLP反应体系.结果表明,用于酶切的基因组DNA以00ng为宜,预扩增产物的稀释倍数以0倍为宜,选择性扩增引物的选择性碱基的数目以"2+"组合为宜.采用优化好的体系,引物组合P12M12在供试材料中共扩增出稳定清晰的带纹5条,其中多态性带4条.

Abstract: AFLP (Amplified Fragment Length Polymorphism) analysis system was established with 24 cucumber self-bred lines. Several factors, include the quantity of genome DNA for cutting, the diluted times of pre-amplifications and the number of selective nucleotide in primer, were optimized. With the optimized system, the AFLP fingerprinting of 24 cucumber materials amplified by primer combination P12M12 have been obtained, which had 35 clear bands, and 4 of them were polymorphic.
