蓖麻矮化相关基因RcDof的克隆及分析

摘要: 为研究RcDof基因在蓖麻矮化中的作用及生物学功能,提取蓖麻生长跃变期茎尖中基因组DNA,根据蓖麻中已获得的锌指蛋白基因片段设计引物,采用RACE技术克隆得到该基因,基因完整阅读框全长为924 bp,可编码307个氨基酸,为C2C2型锌指蛋白,预测蛋白质分子量为34.020 9 kDa,等电点为9.23。二级结构预测表明α螺旋占1.63%,β折叠占1.30%,其他无规则卷曲占97.07%,为外释放蛋白。亚细胞定位于细胞核中,对RcDof基因的生物信息学分析为进一步研究其在蓖麻矮化过程中的作用机制和功能特征提供理论依据。

Abstract: In order to research the features and biological function of RcDof gene in dwarf castor,stem tip genome DNA was extracted in growth climacteric age.Primers were designed according to obtained Zinc finger protein gene fragment.Using RACE technology,the complete full-length open reading frame of 924 bp was cloned,encoding 307 amino acids,belonged to C2C2 zinc finger protein,predicted protein molecular weight was 34.020 9 kDa,isoelectric point was 9.23.The predicted secondary structure α helix accounted for 1.63%,β fold accounted for 1.30%,other non-random coil accounted 97.07%,belonged to outer release proteins.Subcellular localization showed that the RcDof gene located in the nucleus.The bioinformatics analysis of RcDof gene will lay the foundations for the further study on its action mechanism and function characteristics in the castor dwarfing process.
