高邮鸭主要免疫器官的表型发育及其与TLRs表达的相关性分析

Development of Immune Tissue and Correlation Analysis of TLRs Expression in Gaoyou Duck

摘要: Toll样受体(TLRs)在机体天然免疫中发挥重要作用。为分析TLRs在鸭主要免疫器官中的表达情况,在测定和分析高邮鸭免疫器官发育的基础上,荧光定量PCR法检测了高邮鸭TLR1TLR2TLR4TLR5TLR1/2/4/5)4种TLR mRNA基因在免疫器官(胸腺、脾脏和法氏囊)中的相对表达情况,并进行相关性分析。结果表明,TLR1/2/4/5 mRNA在不同发育时间段都有表达,但不同TLR基因在不同免疫组织中表达模式存在差异,总体来讲,胸腺中TLR1/2/4/5 mRNA在1周龄时表达水平较高,脾脏中TLR1/2/5 mRNA在8周龄时表达量显著高于其他周龄,法氏囊中TLR1/5在8周龄时表达量较高。相关性分析结果展示:免疫器官绝对增重和体重发育呈现高相关系数;TLR1/2/4/5 mRNA的表达水平和体重发育相关系数较高,大多高于TLRs mRNA表达水平与免疫器官增重的相关系数;胸腺组织中TLR1/2/4/5 mRNA表达量两两相关系数值相对较高。研究结果可以揭示TLRs在机体及其免疫器官发育过程中的表达情况,为进一步分析和揭示其在免疫抗感染过程中的作用奠定基础。

Abstract: TLRs play an important role in natural immunity.Development of body, thymus, spleen, bursa fabriciu were measured, and the expression levels of TLR1, TLR2, TLR4, TLR5 (TLR1/2/4/5)were detected in these immune tissues by fluorogenic qRT-PCR in Gaoyou duck. The results revealed that the expressions of TLR1/2/4/5 mRNA were detected and showed different pattern in thymus, spleen, bursa fabricius during production period (8 weeks). Expression of TLR1/2/4/5 in thymus at 1 week was higher than those in other weeks, and the higher expressions of TLR1/2/5 mRNA in spleen were showed at 8 weeks, expression of TLR1/5 in bursa fabricius was higher at 8 weeks. Analysis of correlation revealed that there were high index between body weight and immune tissue weight, while the index of expression of TLR1/2/4/5 mRNA and weights of immune tissue were less than the index of expression of TLR1/2/4/5 mRNA and body weight. The expression of TLR1/2/4/5 mRNA in thymus exhibited relatively high correlated index with each other. In conclude, these results can reveal expression pattern of TLR1/2/4/5 during the process of immune organ development.
