一种简单高质量的黄伞菌总RNA分离提取方法

A Simple and High Quality of Total RNA Extraction Method for Pholiota adiposa

摘要: 为了从转录组水平研究黄伞菌的抗肿瘤等药用价值,需要提取分离得到高纯度、高质量的黄伞菌RNA样品。利用液氮研磨TRIzol一步法提取黄伞菌总RNA,并且对经过和未经液氮研磨的黄伞菌RNA提取产物进行了质量检测和统计对比分析。检测结果显示,经液氮研磨样品RNA浓度远远高于未经研磨样品,OD260/280都约等于2,说明其纯度很高。同时,3个重复样品25S和18S亚基条带荧光强度比值分别为1.8,1.9,1.9,接近于2,证明RNA完整性较好,Agilent 2100的RIN值检测结果分别为9.1,8.7,9.3,均远大于6.8标准,进一步佐证了其完整性。所以,液氮研磨TRIzol一步法是非常适用于黄伞菌总RNA高质量提取的简单且高效的方法。

Abstract: Analyzing the function of RNA via RNA-seq technique is a hotspot research currently.In order to study the medicinal value of Pholiota adiposa,such as antitumor mechanism research,on the level of the transcriptome,one need extraction and separation of high purity,high quality Pholiota adiposa RNA samples. This research utilize TRIzol one-step method with liquid nitrogen grinding to extract Pholiota adiposa total RNA.Subsequently,one compared differences of the product between with our method and without. Following,the results are evaluated and processed statistical analysis. Results showed the extracted RNA concentration of the samples via our method was far higher than that without and display a high purity(OD260/280=2). At the same time,agarose electrophoresis experiment exhibited that all fluorescence intensity of three samples was approximately equal to 2,respectively 1.8,1.9,1.9,and all RIN value of three samples via Agilent 2100 are further greater than the standard of 6.8,respectively 9.1,8.7,9.3,indicating that the extracted RNA products have perfect integrity. As a result,TRIzol one-step method with nitrogen grinding is a simple,efficient way and very suitable for extracting the total RNA of Pholiota adiposa.
