百合斑驳病毒大连分离物基因组全序列测定及其分析

Lily Mottle Virus Genome of Dalian Isolates Sequencing and Its Analysis

摘要: 根据GenBank上已登录的百合斑驳病毒(LMoV)基因组序列设计7对交叉重叠的引物,利用RT-PCR技术对铁炮百合白天堂进行检测,并经克隆测序获得百合斑驳病毒大连分离物(LMoV-DL)基因组全序列。序列分析表明:LMoV基因组由长度为9648bp的线状单链正义RNA组成,包括一个poly(A)尾巴,编码一个由3096个氨基酸组成的分子量为351kDa的多聚蛋白。同时多聚蛋白系统发育分析说明LMoV隶属于Potyvirus属一个较大的分支,在进化关系上与BYMV和ClYVV最近。

Abstract: According to Lily mottle virus (LMoV) genome sequence logged in the GenBank,we seven pairs of overlapping primers,and use RT-PCR technique to detect Lilium Longiflorum White Heaven,then obtain the whole-genome sequences of Lily mottle virus of Dalian isolates (LMoV-DL) by cloning and sequencing. Through the sequence analysis indicates that: LMoV genome is the linear positive-sense single-stranded RNA which is constituted with the length of 9648 by bp comprising a poly (A) tail and encoding a molecular weight of 351 kDa poly-protein which is composed of 3096 amino acids. Poly-protein phylogeny analysis that the LMoV is a large branch of Potyvirus and close to BYMV and C1YVV on the evolutionary relationship.
