一种高效提取植物不同组织RNA的方法

A Method for Effective Isolation of Total RNA from Different Plant Tissues

摘要: 为了提高植物不同组织提取质量和效率,降低成本,在热硼酸法的基础上探索出一种适于植物不同组织总RNA提取方法。利用该方法从植物根、茎、叶、不同成熟期果实、不同成熟期种子和不同成熟期果柄中提取的总RNA经紫外分光光度计和琼脂糖凝胶电泳分析证明具有较高的纯度和完整性。所得RNA的A260/A230大于2.3,A260/A280为1.8~2.0,并且RNA产量高,可以满足RT-PCR和Real time RT-PCR的实验需要。该方法能够满足一般分子生物学下游操作的要求,是一种理想的植物不同组织总RNA提取方法。

Abstract: In order to improve the quality and efficiency,reduce the cost of total RNA isolation from different plant tissues A method for effective isolation of total RNA from different plant tissues was invented based on hot borate method.The total RNA isolating from plant root,stem,leaf,fruits of different ripening stages,seeds of different ripening stages,pedicle of different ripening stages using this method had high purity and integrality detected by ultraviolet spectrophotometer and agarose gel electrophoresis.The A260/A230 of yielded total RNA was more than 2.3 while A260/A280 ranged from 1.8 to 2.0.The RNA can be applied to RT-PCR and Real time RT-PCR.This method can meet with demands of down stream experiment of common molecular biology and is an ideal method of plant total RNA extraction.
