高抗体水平肉种鸡H9N2的分离鉴定及HA基因序列分析

Isolation,Identification and Molecular Characterization of a H9N2 Isolate from a Broiler Flock in High Antibody Level Against H9N2

摘要: 从抗体水平较高、产蛋下降的种鸡群分离到一株具有血凝特点的病毒,经HI交叉抑制试验证实为H9N2病毒,命名A/Chicken/Shandong/LY/08,病毒对SPF鸡无致病性,IVPI为0。对其HA基因进行克隆测序,HA基因氨基酸裂解位点的基序为RSSR↓GL,与弱毒基序一致。A/chicken/Shandong/LY/08分离毒株与GenBank上发表的山东参考株HA基因的核苷酸之间的同源性为95.2%~99.6%,氨基酸序列之间的同源性为95.4%~99.5%。与国外A/ck/Korea/ms96/96、A/Parakeet/chiba/1/97和A/turkey/Wisconsin/1/1966 HA基因核苷酸序列同源性为82.6%~91.6%,氨基酸序列同源性为87.9%~92.2%。系统进化树分析表明,该毒株与近年来国内流行的H9N2禽流感毒株类似。

Abstract: A newly H9N2 isolate named A/Chicken/Shandong/LY/08 was collected from a broiler flock in high antibody level against H9N2.The biological test showed that the isolate A/Chicken/Shandong/LY/08 exhibited the IVPI value with 0, and the isolate shared a lentogenic motif 335RSSR↓GL340 at the HA protein cleavage site,which indicated that A/Chicken/Shandong/LY/08 was lentogenic.The HA gene nucleotides ( amino acids) homologies are 95.2%-99.6%( 95.4%-99.5%) between the isolate A/Chicken/Shandong/LY/08 and the Shandong H9N2 isolates.The HA gene nucleotides( amino acids) homologies are 82.6%-91.6%( 87.9%-92.2%) between the isolate A/Chicken/Shandong/LY/08 and A/ck/Korea/ms96/96,A/Parakeet/chiba/1/97,A/turkey/Wisconsin/1/1966 H9N2 isolates.Phylogenetic analysis of HA gene showed that the A/Chicken/Shandong/LY/08 was similar to that of the H9N2 isolates in China.
