摘要: 采用因素轮换试验设计和均匀试验设计对现有枯草芽孢杆菌BE-91菌株的发酵工艺进行系统研究, 获得木聚糖酶高产的优化发酵工艺为:国产胰蛋白胨1.5%, 国产酵母膏0.05%, 牛肉膏0.3%, KH2PO40.2%, MgSO40.03%, NaCl 0.5%, 吐温80 0.4%, 起始pH9.2, 摇瓶装液量150 mL/500 mL, 接种量3.5%, 转速180 r/min, 发酵温度37℃。在优化发酵工艺条件下培养BE-91菌株8 h, 发酵液木聚糖酶活力达到423.24 U/mL, 是优化前酶活力的1.5倍。BE-91菌株在国内已报道菌株中具有发酵周期短、耐热酸性木聚糖酶活力高的特点。

Abstract: The fermentation process of high yield xylanase strain(Bacillus subtilis)BE-91 was optimized by One-Factor-at-a-Time Experimentation Method and Uniform Design Method. The optimal fermentation process were as follows: trypton (made in China) 1.5 %, yeast extract (made in China) 0.05%, beef powder 0.3%, KH2PO4 0.2%, MgSO4 0.03%, NaCl 0.5%, tween 80 0.4%, initial pH 9.2, shake flask capacity 150 mL/500 mL, inoculure concentration 3.5%, stirring rate 180 r/min, temperature 37℃. Under the optimal fermentation process, xylanase activity was up to 423.24 U/mL after 8 h of cultivation, 1.5 times in comparison with the original fermentation process. So the strain BE-91 had advantages of short fermentation period and high thermostable acid xylanase activity in China.
