摘要: 为探究CTAB方法提取植物DNA中影响DNA产量和质量的诸多因素, 以豫花15号花生嫩叶为材料, 采用酚氯仿、1 0000 r/min、氯仿、不剪口、氯仿一步、酚氯仿一步、不摇30 min 7种不同的处理方法纯化其基因组DNA, 并通过外观、紫外分光光度法和琼脂糖凝胶电泳对所提取的DNA样品进行检测, 比较不同因素对DNA提取质量的影响.在充分考虑得率、蛋白质含量及DNA分子完整性等指数后, 总结出酚氯仿纯化豫花15号花生叶片DNA的最适方法。

Abstract: To explore the factors on CTAB method of plant DNA extraction that affect the quantity and quality, seven methods phenol-chloroform, 10 000 r/min, chloroform, no cutting on peptips, one step with chloroform, one step with phenol, no shaking for 30 min have been used to extract genomie DNA from peanut leaves of Arachis hypogaea. The DNA samples obtained by above methods were examed by appearance, agarose gel electrophoresis and ultraviolet spectrophotometry. Based on the comparative analysis of yield and quality of the DNA samples by different methods, consideration all index of yield, protein content and the integrity of DNA molecules, the phenol-chloroform method was found to be the most suitable for extraction of genomic DNA from leaves of A rachis hypogaea.
