朝鲜碱茅BADH基因3'端及5'端部分序列的扩增

The Amplification of BADH Gene 3'and 5'End from Puccinellia chinampoensis

摘要: 为探索朝鲜碱茅的耐盐,耐寒和耐旱的抗性机理,从已知禾本科植物朝鲜碱茅甜菜碱醛脱氢酶(BADH)基因的一段保守区序列,以此序列为研究基础,结合其他禾本科植物的BADH基因保守区序列设计引物,扩增出朝鲜碱茅BADH基因3'端序列及'端部分序列共102 bp.经BLAST比对,结果表明,获得到的朝鲜碱茅BADH基因序列与羊草和大麦的BADH基因的同源性达到89%,含有保守的十肽序列和其后29位与酶功能有关的Cys.

Abstract: Part of conservative sequence of BADH from Puccinellia chinampoensis has been already known and was used as the basis of the research when specific primer was designed. The BADH gene of Puccinellia chinampoensis was cloned by SMART RACE. These two fragments were assembled and the result showed that we obtained the 3′ end and part of 5′ end. The sequence was 1 502 bp in length,its nucleotide sequence share 89 % with the cDNA of BADH from Ley2 mus chinensis and Hordeum brevisubulatum. Its deduced amino acid sequence contains the conserved domain sequence VT LELGGKSP of BADH and the Cys associated with aldehyde dehydrogenase function.
