摘要： 为阐明BMPRIB基因在牛的卵泡发育和分化中的作用及其分子机理，首先根据其他物种BMPRIB基因的保守序列设计特异性引物，从蒙古牛卵巢中提取总RNA，采用RT-PCR技术扩增出蒙古牛BMPRIB cDNA序列；将此片段克隆到pGM-T载体中，提取重组质粒，经PCR鉴定和DNA序列测定分析验证；符合BMP家族基因结构特征，然后根据此序列构建cRNA探针，利用原位杂交技术检测牛卵巢BMPRIB基因 mRNA的表达情况。原位杂交结果显示，牛BMPRIB基因，在初级卵泡和次级卵泡早期表达，在颗粒细胞中表达，在次级卵泡晚期也有表达。

Abstract: Specific primers was designed by conserver sequence of the other animals,total RNA was extracted from ovary of cow and the cDNA encoding cowBMPRIBwas amplified by the reverse transcription PCR(RT-PCR),then the purified RT-PCR product was cloned into PMD18-T vector.The results of PCR analysis of recombinant plasmid and DNA sequencing demonstrated that the cowBMPRIBwas belong to the family of bone morphogenetic protein,BMPRIBmRNA was specialized expressed in cow fore primary and secondary follicle and follicle granulose cell,and expressed terminal secondary follicle by using situ hybridization.
