摘要: 以IBH120鸡胚毒适应成纤维细胞(CEF)单层,培养出IBH120细胞弱毒株。电镜观察,可见典型的冠状病毒粒子。IBH120细胞弱毒株经细菌、霉菌、支原体、鸡白血病病毒检测均为阴性,且具有IB病毒特异性。CEF单层接毒后48h左右病毒增殖达到高峰。随着细胞传代次数的增加,C4~C10代H120株细胞毒的毒价(TCID50)逐步增高而趋于稳定,C4~C10代细胞毒的TCID50稳定在107.35~8.0mL之间,而C5~C10代IBH120细胞毒的EID50呈下降趋势,变动范围在107.3~106.5mL。以103.5TCID50/只和104.5TCID50/只的剂量,C4,C8,C10代毒分别滴鼻接种3日龄敏感鸡,免疫鸡的攻毒保护率为80%~100%,病毒分离回收率分别为20%和0。

Abstract: Cellular attenuated strain of infection bronchitis H120 strain(IBH120)was cultured successfully with virus which cultured on the chicken embryo.Atypical Coronary viral partical were observed by electric microscope.IBH120 cellular attenuated strains were negative after examination bacterium,mildew,mycoplasma,chicken leukemia virus,moreover,they speciticity of IB virus.Virus proliferate could reach peak after 48 h.When CEF cells infected with IBH120,TCID50 of C4-C10 generation of H120 strain cellular attenuated virus rose gradually and stabilized with increasing generation.TCID50 of C4-C10 generations were between 107.35 and 108.0.but EID50 of C5-C10generation were degressive trend.The variational range was between 107.3 and 106.5.Theimmunogenicity of IBH120 cellular attenuated virus was checked.The result demonstrated that three days old chickens were inoculated in nose with 103.5 TCID50 and 104.5 TCID50 of C4,h,C10 generation virus in each chicken respectively,their protection rate was between 80% and 100%.The virus separation recovery rate was 20% and 0 respectivelly.
