摘要: 采用两阶段培养法研究不同培养基、激素的种类、浓度和各种营养成分对愈伤组织生长及紫草素形成的影响.结果表明,改良B培养基为适宜的营养生长培养基.细胞分裂素和生长素均为紫草素形成所必需的.6-BA 1.0 mg*L-1,IAA 0.1 mg*L-1为较佳配比.生长培养基中CaCl2浓度为20 mg*L-1,MgSO4为00 mg*L-1时,愈伤组织生长和紫草素形成均较佳.紫草素形成培养基中CuSO4为M9浓度的3倍时,紫草素产量较高.紫草素形成培养基中添加VB2及更高的FeSO4对紫草素产量的提高没有效果.研究过程中优化了紫草细胞培养形成紫草素的方法,为建立完整的组培生产紫草素体系提供了基础数据.

Abstract: Tw o-stage culture method w as used to study the ef fect of types and concentration of hormone, effect of types of culture medium, and nut rit ion on shikonin format ion. The result s show that both cytokinin and aux in are needed in shikonin formation. 6-BA 110 mg#L-1 and IAA 011 mg#L-1 is suitable in shikonin format ion medium. B5 medium is a better medium for cell growth. B5 w ith 250 mg#L-1 of CaCl2, 500 mg#L-1 of Mg SO4 are bet ter for both growth and shikonin format ion. M9 w ith three t imes of concentration of CuSO4 are bet ter choice for shikonin format ion. Addition of VB2 and more FeSO4 has no posit ive ef fect on shikonin format ion.
