摘要: 用二倍体多年生类玉米、玉米自交系330及杂种F1(玉米自交系330×二倍体多年生类玉米)的幼雄穗进行离体培养,获得高频率的植株再生。幼雄穗培养在含2mg/16-BA的MS培养基上直接产生不定芽。在这一培养基上继代培养,建立了不定芽的快速无性繁殖系。不定芽转移到生根培养基(MS附加0.5mg/lNNA,300mg/l水解乳蛋白和1000mg/l活性碳),非常容易生根。幼雄穗的发育阶段和植物激素的配比,对培养物的成功建立是非常重要的。本文还讨论了这一技术的应用。

Abstract: Plants were regenerated with high frequency from immature tassels of Zea mays L.(maize inbred 330),Zea diplopetennis,and Zea maysZea×Zea diplopennis hybrid cutured in vitro.Adventitious buds and shoots were directly developed in the primary culture from the immature tassels cultured on the medium MS containing 2 mg/1 6-BA.Subcultured on the same medium,rapid,clonal bud and shoot propagation was obtained.The buds and shoots were very easy of rooting by transferring them to root-inducing medium,MS containing 0.5 mg/1 NAA,300 mg/1 lactalbumin hydrolyzate(LH),and 1000 mg/1 activated charcoal.The developmental stage of immature tassels and the combination of phytohormones were most important for successful culture establishment.The application of this technique was discussed.
