摘要: 蓖麻是一种重要的油料作物,种子中含有毒蛋白,蓖麻毒蛋白具有相当明显的抗肿瘤作用。以2129蓖麻品系不同发育时间的鲜种子为材料,利用磷酸盐提取法、BCA试剂盒法和UVwin5紫外V5.1.0、BandScan蛋白定量分析、Excel、IBM SPSS Statistics软件,研究种子中毒蛋白的动态积累过程。结果表明:不同发育时间蓖麻鲜种子中,毒蛋白的含量随着发育时间的增加而逐渐在发育完全成熟即60 d时达到最大;毒蛋白含量与粗蛋白含量、种仁质量、种子质量分别呈极显著正相关、显著正相关和正相关关系。基于种子中毒蛋白含量将蓖麻鲜种子的发育时间分为3类:第Ⅰ类包括30,40,50,60 d,这些发育时间蓖麻鲜种子中蓖麻毒蛋白含量较高;第Ⅱ类只包括10 d,毒蛋白含量较低;第Ⅲ类只包括20 d,毒蛋白含量中等。不同发育时间蓖麻鲜种子中毒蛋白的动态积累过程是,在种子发育10~20,20~30 d时迅速积累,30~60 d时积累速度较慢,完全成熟即60 d时毒蛋白含量达到最大。

Abstract: Castor is an important oil crops,seeds containing ricin,which have an quite obviously effect on anti-tumor.In this experiment,we used 2129 castor fresh seed which at different development time for the study,using phosphate extraction method,BCA kit method,UVwin5,ultravioletV5.1.0,BandScan protein quantitative analysis and Excel,IBM SPSS Statistics software to research dynamic accumulation process of ricin in seed.The results were as follows:ricin content of fresh castor seeds at different developmental time increases with development time,and gradually reached the maximum in 60 d when fully mature;ricin content and crude protein content was high-significant positive correlation,with quality of seeds was significant positive correlation,and seed quality were positive correlation.Based on seed ricin content fresh castor seed development time will be divided into three categories:ClassⅠincluding 30,40,50,60 d,these castor seeds ricin content was higher,typeⅡincluded only 10 d,low ricin content;class Ⅲ included only 20 d,moderate ricin.The conclusion is as follows:dynamic accumulation process of ricin in fresh castor seeds at different developmental time is rapidly accumulated during seed development 10-20,20-30 d,then 30-60 d became slow,ricin content reaches the maximum when 60 d fully ripe.
