摘要: 为了获得具有标记的猪瘟病毒Erns蛋白,通过基因工程操作,使猪瘟病毒Erns基因与绿色荧光蛋白基因(GFP)融合,用Bac-to-Bac杆状病毒表达系统在昆虫细胞中高效表达了ErnsGFP融合蛋白,经WesternBlot和荧光显微镜观察证实,表达产物分子量正确,且发出易于检测的绿色荧光,具有标记的Erns蛋白的获得为进一步研究Erns蛋白结构和功能奠定了基础。

Abstract: To obtain labeled Erns protein of classical swine fever virusthe coding region of Erns and green fluores-cent protein ( GFP) was fused by genetic engineering technique,and expressed in cells according to the manual ofBacoac baculovirus expression system. The results of Western Blot analysis and fluorescence microscopy show thatthe fused proteins were successfully expressed with spontaneous green fluorescent. The established method of obtainingthe labeled protein will be helpful for further exploring the structure and function of Erns protein.
