五家渠尖缘螺线粒体基因组测序分析

Complete Mitochondrial Genome Sequence of Oxyloma wujiaquensis Li, Guo & Zhang, 2017(Gastropoda:Succineidae)

摘要: 为了获得五家渠尖缘螺线粒体基因组全序列,分析其组成特征,探讨其与同科的腐败琥珀螺以及肺螺亚纲相关类群之间的系统发育关系,运用Illumina Hiseq 2000高通量和Sanger测序技术以及MITOS在线注释及Genious R11.0等软件,对五家渠尖缘螺线粒体基因组序列进行了测序与分析。结果表明,五家渠尖缘螺线粒体全基因组序列长度为14 086 bp (GenBank登录号:MT670402),AT含量为75.64%,GC含量为24.36%。全基因组序列由13个蛋白质编码基因,22个tRNA基因,2个rRNA基因及1个非编码区组成。13个蛋白质编码基因使用ATN、TTN等为起始密码子,使用TAN、CAT、T等为终止密码子。22个tRNA基因中tRNASer1和tRNASer2缺失DHU臂。A+T富集区位于COX1与tRNAVal之间,长度为42 bp。五家渠尖缘螺和同科的腐败琥珀螺线粒体基因组基因排列顺序和结构基本相似。从GenBank上下载有关肺螺亚纲及后鳃亚纲的部分种类线粒体全基因序列,构建基于线粒体基因组序列的系统进化树,并对琥珀螺科种类进行系统发育分析。结果显示,五家渠尖缘螺与腐败琥珀螺聚在一起,有较近的亲缘关系。目前,在GenBank有关琥珀螺科线粒体全基因组序列的信息非常少,本研究丰富琥珀螺科种类的基因序列信息,为将来的琥珀螺种类的系统发育分析、资源保护及遗传多样性等研究提供基础数据。

Abstract: In order to obtain the complete sequence of the mitochondrial genome of Oxyloma wujiaquensis Li, Guo & Zhang, 2017, analyze its composition characteristics, and explore the phylogenetic relationship between it and related groups in the family Succineidae and class Gastropoda, using Illumina Hiseq 2000 high-throughput and Sanger sequencing technology, MITOS online annotation and Genious R11.0 software, sequenced and analyzed the mitochondrial genome sequence of Oxyloma wujiaquensis. The results showed that the entire mitochondrial genome sequence was 14 086 bp (GenBank accession number:MT670402) with 75.64% AT and 24.36% GC, containing 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes and non-coding regions.13 protein-coding genes used ATN, TTN as start codons and TAN, CAT and T as stop codons. In 22 tRNA genes, tRNASer1 and tRNASer2 lacked DHU arm. The A+T enrichment region was 42 bp, located between COX1 and tRNAVal. The gene sequences and structures of mitochondrial genome of Oxyloma wujiaquensis and Succinea putris Linneaus, 1758 which belonged to the same family Succineidae were same basically. The mitochondrial genome sequence information of some species of Pulmonata and Opisthobranchia, construct a phylogenetic tree based on the mitochondrial genome sequence, and conduct phylogenetic analysis on the species of Succineidae. The results showed that these two species were gathered together, showing a close relationship. At present, there is very little information about the mitochondrial genome sequence of Succineidae in GenBank. This study will enrich the gene sequence information of Succineidae species and provide future research on the phylogenetic analysis, resource protection and genetic diversity of Succineidae provide basic data.
