黄野螟谷胱甘肽硫转移酶Sigma 1基因的鉴定及表达模式

Characterization and Expression Pattern of Glutathione S-transferase Sigma 1 Gene from Heortia vitessoides

摘要: 为探究谷胱甘肽硫转移酶Sigma家族中Sigma 1基因的分子特征,从黄野螟成虫转录组文库中鉴定获得了GST Sigma 1基因全长cDNA,命名为HvGSTs1(GenBank:MF521977)。并对该基因进行生物信息学分析,并使用RT-qPCR对HvGSTs1在其不同发育阶段、幼虫不同部位及成虫不同部位的相对表达量进行检测。结果表明,该基因全长1 054 bp,共编码204个氨基酸。序列分析显示,HvGSTs1蛋白氨基酸序列含有N端结构域GST_N_Sigma_like和C端结构域GST_C_Sigma_like等2个结构域,HvGSTs1的氨基酸序列与二化螟同源性最高,为76%。系统发育树分析显示,黄野螟与二化螟处于同一分支。用RT-qPCR分析了HvGSTs1基因的相对表达量,结果显示,HvGSTs1在蛹中的表达量高于其他发育阶段;HvGSTs1在幼虫脂肪体中表达量最高,在中肠表达量最低;HvGSTs1在成虫中腹部和胸部表达量最高,足部表达量最低,各部位均有表达,且有显著差异(P < 0.05)。研究结果为以后深入探讨黄野螟Sigma家族GST的生理功能奠定了基础。

Abstract: To elucidate the molecular characteristics of Sigma 1 gene of glutathione S-transferase sigma family,the gene named HvGSTs1 (GenBank:MF521977)was screened from the transcriptome of the adult of Heortia vitessoides. The gene was analyzed by bioinformatic approaches,and RT-qPCR was used to detect the relative expression of HvGSTs1 in various developmental stages and in different larval and adult tissues. The results showed that HvGSTs1 was 1 054 bp in length,encoding 204 amino acids. Sequence analysis showed that the amino acid sequences of HvGSTs1 protein contained two conserved domains,namely GST_N_Sigma_like and GST_C_Sigma_like. HvGSTs1 showed the highest homology with Chilo suppressalis (76%). Phylogenetic tree built by ClustalX and MEGA 4 indicated H.vitessoides with C.suppressalis were in the same branch. The relative expression levels of HvGSTs1 detected by RT-qPCR indicated that the expression levels of HvGSTs1 in pupae were higher than that in other stage of development. In larvae, HvGSTs1 had the highest expression level in fat body,nevertheless,the lowest in midgut. In adults, HvGSTs1 reached the highest expression level in abdomen and thorax and the expression level in foot was the lowest. The HvGSTs1 were prominently and differently expressed in all of the adult parts(P < 0.05). It lay the foundation for further discussion on physiological functions of Sigma family GST of H.vitessoides.
