抗谷胱甘肽-S-转移酶(GST)单克隆抗体的研制及初步应用

摘要: 以纯化的谷胱甘肽转移酶标签蛋白(GST)免疫Balb/C小鼠,取免疫鼠脾细胞与NS0骨髓瘤细胞按常规方法融合,用纯化的GST经间接ELISA筛选,阳性孔经3次有限稀释法亚克隆,最终获得2株抗GST的单克隆杂交瘤细胞株,分别命名为4B7-E4和4B7-F4。间接ELISA检测4B7-E4、4B7-F4细胞培养上清的效价分别为1∶4 000、1∶3 000,腹水效价分别为1∶3×106和1∶2×106。Western-blot结果表明,这两株单抗均可以特异性地识别GST蛋白和带有GST标签的融合蛋白。

Abstract: The monoclonal antibodies(McAbs) against Glutathione-S-transferase(GST) were prepared in the present work.Balb/C mice were immunized with purified GST and the cell fusion was performed by standard method.The hybridom secreting antibodies were subcloned three times by limiting dilution.Two hybridomas,which were named as 4B7-E4 or 4B7-F4 and secreted McAbs against GST,were picked out by indirect ELISA.The ELISA antibody titers of cell culture supernatant and ascite of 4B7-E4 and 4B7-F4 were 1∶ 4 000 or 1∶ 3 000,and 1∶ 3×106 or 1∶ 2×106 respectively.Western-blot showed that these.McAbs can specifically react with GST and recombinant GST.
