甲状腺素及胰高血糖素对高温条件下体外培养奶牛乳腺上皮细胞热休克蛋白表达的影响

The Effects of Thyroxine and Glucagon on Heat Shock Proteins Expression of Bovine Mammary Epithelial Cells under Heat Circumstance in vitro

摘要: 为阐明甲状腺素及胰高血糖素对高温体外培养奶牛乳腺上皮细胞内热休克蛋白(HSPs)mRNA转录和蛋白表达的影响。本试验对体外培养的奶牛乳腺上皮细胞添加不同浓度的甲状腺素、胰高血糖素,分别于42℃培养12 h。采用RT-qPCR方法检测细胞内HSF-1、HSP27、HSP70和HSP90的mRNA表达丰度,ELISA方法检测HSP27、HSP70和HSP90的蛋白质表达。各浓度的甲状腺素、胰高血糖素均能显著降低高温条件下奶牛乳腺上皮细胞中HSP27、HSP70和HSP90 mRNA表达丰度(P<0.05);各浓度的胰高血糖素能极显著的降低高温条件下HSP27、HSP70和HSP90的蛋白表达(P<0.01),同时各浓度甲状腺素也能极显著的降低HSP27、HSP70的蛋白表达(P<0.01),而仅0.01,0.1,2μmol/L的甲状腺素能极显著的降低HSP90的蛋白表达(P<0.01)。结果提示,甲状腺素及胰高血糖素能明显降低高温条件下HSPs mRNA的转录和蛋白的表达水平。

Abstract: The experiment was conducted to study the effects of thyroxine(T4)or glucagon on heat shock proteins( HSPs)expression of bovine mammary epithelial cells cultured under heat circumstance in vitro.Bovine mamma. ry epithelial cells were cultured with different concentration of T4 or glucagon, which were exposed at 42%as heat treatment model for 12 h.Then mRNA transcript and protein expression were detected by RT—qPCR and ELISA.The resuhs showed as follows:Every experimental concentration of T4 and glucagon can significantly decrease the mRNA transcript level of HSP27, HSP70, HSP90, HSF·1(P<0.05), which ceils cultured at 42。C;The protein expression can also decreased extremely markedly due to cells cultured with different concentration of glucagon at 42%;at the same time, every experimental concentration of T4 can significantly decrease the protein expression of HSP27, HSP70 at 420C(P<0.01), but only 0.01, 0.1, 2 p。mol/L of T4 had the significantly effects on HSP90(P<0.01).In conclusion, T4 and glucagons can decrease HSPs mRNA transcript and protein expression level of cells cultured under heat circumstance.
