为准确了解谷物中木聚糖酶抑制蛋白活性的存在情况,从而进一步分析这种抑制蛋白活性对谷物加工中外加木聚糖酶可能产生的负面影响。选取小麦、大麦、黑麦、绿粒小麦、荞麦、燕麦、玉米、水稻和高粱9种常见的谷物为材料,测定了其籽粒以及籽粒不同部位木聚糖酶抑制蛋白的活性。结果表明,木聚糖酶抑制蛋白活性在小麦、黑麦、绿麦和荞麦中较高,分别为4.27,4.76,4.18,3.61 IA/mg;在大麦、燕麦中较低,分别为0.54,0.87 IA/mg;玉米、水稻和高粱中没有发现木聚糖酶抑制蛋白活性。进一步选取木聚糖酶抑制蛋白活性较高的小麦、黑麦和绿粒小麦,分别测定其种皮、胚和胚乳中的木聚糖酶抑制蛋白活性。结果显示,小麦种皮、胚和胚乳中的木聚糖酶抑制蛋白活性分别是0.06,3.67,4.47 IA/mg;黑麦种皮、胚和胚乳中的木聚糖酶抑制蛋白活性分别是1.70,5.42,5.39 IA/mg;绿粒小麦种皮、胚和胚乳中的木聚糖酶抑制蛋白活性分别是0,5.06,4.57 IA/mg。3种麦类表现出一致的结果,即木聚糖酶抑制蛋白活性在胚和胚乳中基本持平,并且远高于种皮。
To obtain the accuRate infoRmation about the activity of xylanase inhibitoR pRotein in diffeRent ceReals and fuRtheR investigate its potential negative effect on the function of exogenous xylanase used in ceReal pRocessing, the keRnels of 9 commonly planted ceReals including wheat,baRley, Rye,gReen wheat,buckwheat,oats,coRn, Rice and soRghum weRe selected as expeRimental mateRials to study the xylanase inhibitoR pRotein activity and the activity distRibution in diffeRent fRactions. The Results showed that the xylanase inhibitoR pRotein activity in wheat, Rye,gReenwheat, a nd buckwheat( 4. 27,4. 76 4. 18 and 3. 61 IA/mg) was much higheR than in baRley and oats( 0. 54 and 0. 87 IA/mg) . The xylanase inhibitoR pRotein activity weRe not detected in coRn, Rice and soRghum. Then wheat, Rye and gReen wheat with higheR xylanase inhibitoR activity weRe fuRtheR used to deteRmine the distRibution of xylanase inhibitoR pRotein activity in diffeRent fRactions of keRnel. The inhibiting activity in seed coat,embRyo and endospeRm weRe 0. 06,3 . 67 and 4. 47 IA/mg in wheat Respectively,1 . 70,5 . 42 and 5. 39 IA/mg in Rye Respectively,and 0, 5. 06 and 4. 57 IA/mg in gReen wheat Respectively. It was concluded that the xylanase inhibitoR pRotein activity was much higheR in the embRyo and endospeRm than in the seed coat.
